SD/-Met/-Trp Broth (SD/-Met/-Trp medium for short) is a commonly used bacterial culture medium. The medium is widely used in molecular biology and genetic engineering research for breeding bacteria with specific genetic traits.
SD/-Met/-Trp Broth is prepared as follows:
1. Prepare SD medium: Dissolve 6.7g Difco Synthetic Dropout Base powder in 500mL deionized water and adjust pH to 5.8.
2. Add 1mL 20% glucose solution to make the concentration 0.04.
3. add 0.77g SD powder (-Met/-Trp) and fully dissolve.
4. The final volume is 500mL, adjusted to the desired volume by deionized water.
SD/-Met/-Trp medium is used to screen for bacteria with specific genetic traits. This medium lacks the proteins methionine (Met) and tryptophan (Trp), and for bacteria that cannot synthesize these two amino acids on their own, only bacteria that carry the corresponding genes can grow. Therefore, SD/-Met/-Trp medium is often used to select bacteria that carry the expression of foreign proteins, such as recombinant protein expression vectors.
